Blood vessels widen and narrow to control blood flow. In diabetes this control often fails, and the resulting damage underlies many of the disease's complications.

Grants

What they work on

  1. Determine whether hemoglobin alpha in artery-lining endothelial cells becomes glycated in a mouse model of diabetes, including at sites within its putative eNOS-binding domain.
  2. Disrupt the interaction between hemoglobin alpha and eNOS, the nitric oxide–producing enzyme, with pharmacological and genetic approaches, and test how this affects the development of diabetic vascular dysfunction.
  3. Generate two conditional knock-in mouse lines expressing red fluorescent protein–based indicators by using the CRISPR-based RE-CREATING method to replace the GFP indicator in an established mouse strain.
  4. Characterize and validate the red indicator mice by imaging calcium and membrane potential while using pressure myography and sharp microelectrode recordings in blood vessels, then share them with the research community.

More about the research

The research here has two strands. One asks how a protein inside artery walls, hemoglobin alpha, may contribute to vascular damage in diabetes. The other builds new mouse tools for watching activity inside living cells in red light.

Why it matters

In people with diabetes, a damaged hemoglobin alpha–eNOS interaction in artery walls could serve as a new marker of cardiovascular risk. It could also become a target for drugs that protect blood vessels. Separately, red fluorescent indicator mice could give researchers across many fields clearer imaging of calcium and voltage signals in living tissue. They would also allow two indicators to be imaged in the same animal.
