When a newborn's brain is deprived of oxygen and blood flow, an event called hypoxic ischemic (HI) insult, both white matter (the nerve fibers that connect regions) and grey matter (the nerve cell bodies) are damaged. HI injury is a major cause of infant death and long-term disability, and the only effective treatment is therapeutic hypothermia, deliberate cooling of the body.

What they work on

  1. Develop diffusion MRI methods that measure tissue microstructure and water exchange between inside and outside of cells in a neonatal mouse model of hypoxic ischemic injury, and validate them against histology.
  2. Use the new diffusion MRI measures to characterize the extent and severity of brain cell death and to predict which edema (swollen) regions will survive.
  3. Cross-validate the diffusion MRI markers against metabolite and lipid maps of the same injured brains obtained by mass spectrometry imaging.
  4. Apply the validated imaging techniques to mice treated with therapeutic hypothermia to measure its effects on injury progression and elucidate its neuroprotective mechanisms.

More about the research

Animal models are widely used to study how the damage unfolds and how cooling helps. This project builds imaging tools to follow that process in living animals.

Why it matters

Infants with oxygen-deprivation brain injury have only one proven treatment, cooling. Imaging measures that reveal cell death and predict whether swollen tissue will recover could help researchers design better interventions and track treatment response in preclinical studies, and may clarify how therapeutic hypothermia protects the newborn brain.
